Depends what you want to do. For some cultural experience go to St Kilda or Carlton
Personally I like around Albert Park...it's a beautiful area close to the beach with lots of gorgeous historical architecture, but just a quick tram-ride into the city.
South Wharf area is a great place to stay. Close to the city tourist sites, and accessible by public transport.
Darling Towers @ 233 Collins Street.. Stayed at the serviced apartment there in the Summer of '06. It is a wonderful place central to all sight-seeing points and close to all public transport too!
If I was spending a weekend I would stay in around south bank lots going on an easy access to markets in the north or Saint kilda etc
When my friends from overseas visited Melbourne, I asked them to stay in the CBD: the Sky Bus is only 20 minutes to and fro the airport, there are trams and trains going to all parts of Melbourne from the city, plus there are so many amazing spots within the CBD itself - great coffee, talented buskers, delicious food and events aplenty.
Sofitel in Southbank is a good candidate because of the central location, good services, close proximity to eateries and shopping centres and best of all for the in-house restaurant known as The Melba, Melbourne's best buffet restaurant.
